Observability and pharmacological calibration in psychoactive ethnobotany: Admixture selection and the discovery of Ayahuasca

Abstract

Background: Traditional pharmacopoeias, including psychoactive plant traditions, contain pharmacologically active compounds at rates far above chance. The Amazonian ayahuasca complex — a brew requiring co-administration of an N,N-dimethyltryptamine (DMT) source with a monoamine-oxidase inhibitor (MAO-I) — is the paradigm case: indigenous practitioners arrived at a highly specific pharmacological pairing navigating a flora of 14003 species. We apply the observability gradient framework to evaluate when traditional pharmacological knowledge tracks empirical reality.

Methods: Three tests: (i) cross-cultural comparison of ceremony versus pharmacokinetic effect duration across eleven independent traditions spanning five continents and seven pharmacological classes; (ii) a 118-plant Amazonian admixture catalogue cross-classified by purpose observability and pharmacological validity (70 years of documentation); (iii) an agent-based simulation of guided iterative search for the DMT + MAO-I combination.

Results: Ceremony duration tracks pharmacokinetic duration log-log (Pearson r = 0.977, p = 2.5 × 10⁻⁷, n = 11; slope 1.010). Active admixture plants cluster at the extremes of the purpose-observability distribution while candidate plants concentrate in the middle (χ²(2) = 20.2, p = 4.2 × 10⁻⁵), a bimodal pattern: the relationship between purpose observability and contemporary persistence is non-monotonic. Guided iterative search converges on the DMT + MAO-I target in centuries; pure random search fails within archaeologically plausible bounds.

Conclusions: Traditional psychoactive ethnobotanical knowledge tracks pharmacological reality where outcome-feedback is observable. Observability offers a principled triage criterion for ethnopharmacological follow-up; small samples and single-author admixture coding warrant replication.
